Target Information
RPS6KA5 (MSK1) is a serine/threonine protein kinase essential for regulating various cellular processes. It mediates mitogen or stress-induced phosphorylation of transcription factors such as CREB1, ATF1, RELA, STAT3, and ETV1/ER81. MSK1 contributes to gene activation via histone phosphorylation and regulates inflammatory gene expression. It phosphorylates CREB1 and ATF1 in response to stimuli such as UV-C irradiation, EGF, and anisomycin. MSK1 also modulates RELA transcriptional activity upon TNF stimulation and interacts with the glucocorticoid receptor NR3C1 to repress inflammatory gene expression. In skeletal myoblasts, it phosphorylates RELA at Ser-276 during oxidative stress, and in erythropoietin-stimulated cells, it is required for Ser-727 phosphorylation of STAT3.
