Target Information
The progesterone receptor (PR) is a member of the steroid family of nuclear receptors. It mediates the physiological effects of progesterone, which plays a central role in reproductive events associated with pregnancy. PR exists as two forms: a 94 kDa protein (Form A) and a 120 kDa protein (Form B), produced by alternative translation initiation sites.
PR-B is transcriptionally active, responsible for gene activation related to endometrial maintenance, pregnancy, and ovulation inhibition. PR-A lacks 165 amino acids at the N-terminus, exposing a 140 amino acid inhibitory domain that represses steroid hormone transcriptional activity.
In its inactive state, PR forms a multiprotein complex including heat shock proteins and immunophilins. Upon progesterone binding, the receptor undergoes conformational change, dimerizes, and binds to progesterone response elements (PRE), activating transcription. Null mutations in the PGR gene cause reproductive abnormalities.
