Product Specific Information

This antibody detects endogenous levels of p53 only when phosphorylated at Serine 315.

Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nutrient deprivation.
p53 activation leads to transcription of genes such as p21waf1 involved in G1 or G2/M arrest. Structurally, p53 includes an N-terminal transactivation domain, central DNA-binding domain, oligomerization domain, and C-terminal regulatory domain.

Phosphorylation at Ser15 is critical for p53 activation and stabilization, while phosphorylation at Ser392 is associated with tumor formation. p53 also plays roles in aging, oxidative stress, and learning and memory processes.
In normal cells, p53 levels are low, but in transformed cell lines, it is highly expressed and contributes to malignancy. Mutations in p53 disrupt DNA binding and tumor suppressor function, occurring both somatically and germline (e.g., Li-Fraumeni syndrome).
