Target Information
The 6x His tag is a synthetic oligopeptide consisting of six consecutive histidine residues (HHHHHH). It is commonly expressed at either the N- or C-terminal regions of recombinant proteins to enable isolation or purification via immobilized metal affinity chromatography (IMAC).
Epitope tagging involves fusing a known epitope to a recombinant protein using genetic engineering, allowing detection of proteins lacking specific antibodies. His-tagged proteins are purified using metal chelate chromatography due to the strong and selective binding affinity of histidine clusters to metal ions such as Ni²⁺.
His tag-specific antibodies facilitate detection and co-immunoprecipitation of His-tagged proteins.
