His 태그는 재조합 단백질의 N- 또는 C-말단에 부착되어 금속 친화성 크로마토그래피(IMAC)를 통해 분리 및 정제를 가능하게 합니다. 에피토프 태깅(epitope tagging)은 알려진 에피토프를 재조합 단백질에 유전공학적으로 융합하여 항체가 없는 단백질도 검출할 수 있게 하는 기술입니다. His 태그 단백질은 히스티딘 클러스터의 금속 결합 특성을 이용해 Ni²⁺-NTA 흡착제를 통한 친화 크로마토그래피 정제에 사용됩니다. His 태그 특이 항체는 태그 단백질의 검출 및 공동면역침전(co-immunoprecipitation)에 활용됩니다.
