Casein kinase I (CKI) and casein kinase II (CKII) are serine/threonine protein kinase families found in all eukaryotes.
CKI family members (α, γ, ε, δ) are involved in cytoplasmic and nuclear processes such as DNA replication and repair.
CKII typically forms a tetrameric complex (α₂β₂ or αα′β₂), with the α catalytic subunit activated by the β regulatory subunit, which undergoes autophosphorylation.
CKII phosphorylates over 100 substrates, including nuclear oncoproteins, transcription factors, and enzymes involved in DNA metabolism.
