• For immunohistochemistry (PFA fixed): incubate antibody for 30 minutes at room temperature.
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

CD34 is a highly glycosylated monomeric surface protein (111–115 kDa) expressed on various stem cell populations. It functions as a stem cell marker, though its expression on human hematopoietic stem cells is reversible. CD34 may participate in receptor-mediated endocytosis and regulate adhesion, differentiation, and proliferation of hematopoietic stem and progenitor cells.

It likely mediates attachment of stem cells to the bone marrow extracellular matrix or stromal cells and may act as a scaffold for glycans that bind to lectins. CD34 is also thought to present carbohydrate ligands to selectins. The intracellular domain can be phosphorylated by protein kinase C, suggesting a role in signal transduction.
Diseases associated with CD34 dysfunction include dermatofibrosarcoma and neurofibroma.
