• Immunohistochemistry (PFA fixed): Incubate antibody for 30 minutes at room temperature.
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

CD34 is a highly glycosylated monomeric surface protein (111–115 kDa) expressed on various stem cell populations. It serves as a stem cell marker and may function as a surface receptor involved in receptor-mediated endocytosis, adhesion, differentiation, and proliferation of hematopoietic stem cells and progenitors.
CD34 expression represents a specific state of hematopoietic development with altered adhesion properties. It may mediate attachment of stem cells to the bone marrow extracellular matrix or stromal cells, and act as a scaffold for lineage-specific glycans. CD34 also plays a role in presenting carbohydrate ligands to selectins.
The intracellular chain of CD34 is phosphorylated by activated protein kinase C, suggesting a role in signal transduction.
Diseases associated with CD34 dysfunction include dermatofibrosarcoma and neurofibroma.
