Product Specific Information

The OKT4 monoclonal antibody reacts with human CD4, a 59 kDa cell surface glycoprotein expressed by thymocytes, T-helper cells, and monocytes. CD4 serves as a receptor for HIV. The OKT4 and RPA-T4 antibodies recognize different epitopes and do not cross-block each other.

This RPA-T4 antibody is pre-diluted and validated for flow cytometric analysis of normal human peripheral blood cells. Recommended usage is 5 µL (0.125 µg) per test, defined as the amount required to stain a 100 µL sample containing 10⁵–10⁸ cells.

Super Bright 780 is a tandem dye excited by the violet laser line (405 nm) and emits at 780 nm. Use a 780/60 bandpass filter for optimal detection.

When using multiple Super Bright dye-conjugated antibodies, employ Super Bright Complete Staining Buffer (Product # SB-4401-42) to minimize non-specific polymer interactions.

Compensation Notes:
Higher compensation values may occur in the violet 450/50 channel when using UltraComp eBeads (Product # 01-2222-42) compared to single-color stained cells. In such cases, set compensation using cells. Similar observations may occur with AbC Total Antibody Compensation beads (Product # A10497).

Light Sensitivity:
Super Bright 780 is photo-sensitive; protect from light during storage and use.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Target Information

CD4 antigen recognizes MHC class II molecules and acts as a co-receptor for HIV. It is mainly expressed in T-helper lymphocytes but also found in monocytes, macrophages, and dendritic cells. Tissue distribution includes thymus, lymph nodes, tonsils, spleen, and certain non-lymphoid tissues. CD4 contributes to early T-cell activation and may mediate neuronal damage in immune-related CNS diseases. Multiple alternatively spliced transcripts have been identified (RefSeq, July 2017).
