Product Specific Information

The OKT3 monoclonal antibody reacts with an epitope on the epsilon-subunit within the human CD3 complex. It has potent immunosuppressive properties and has been effective in treating renal, heart, and liver allograft rejection. CD3 is expressed by thymocytes and all mature T cells. Crosslinking of TCR initiates intracellular signaling for activation and proliferation.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and tested with human peripheral blood cells at 5 µL (0.5 µg)/test.
Laser: Violet (405 nm); Emission: 780 nm.

Super Bright 780 is a tandem dye excited at 405 nm and emits at 780 nm. Use a 780/60 bandpass filter. Protect from light due to photo-oxidation sensitivity.

Recommended Buffer: Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.
Fixation: Samples can be stored up to 3 days in the dark at 4°C using IC Fixation Buffer or 1-step Fix/Lyse Solution.

Target Information

The CD3 complex transduces antigen-recognition signals in T cells and regulates TCR expression. CD3 subunits (gamma, delta, epsilon, zeta) are encoded on chromosome 11 and mediate signal transduction through tyrosine-based motifs. CD3 is expressed on 68–82% of peripheral blood lymphocytes and 65–85% of thymocytes but not on B or NK cells. Defects in CD3 genes are associated with immunodeficiency.
