TSC2 (Tuberin, Tuberous sclerosis complex, TSC Complex Subunit 2)
TSC2는 종양 억제 인자로 알려져 있으며, 세포 성장 정지 조절 및 스테로이드 수용체 매개 전사 조절에 관여합니다. Hamartin과 복합체를 형성하여 세포질 내에서 샤페론 역할을 할 수 있으며, Ser939 및 Thr1462의 인산화는 Hamartin과의 상호작용을 조절합니다. 이는 phosphoinositide 3-kinase/Akt 경로를 통해 성장 인자에 의해 자극됩니다. 또한 TSC2는 세포 성장 조절 및 소포 수송에 관여하며, RAP1A 및 RAB5의 내재적 GTPase 활성을 촉진합니다. TSC2 유전자 변이는 RAP1A의 지속적 활성화와 관련되어 있으며, 림프관평활근종증(lymphangioleiomyomatosis) 및 TSC2 혈관근지방종(angiomyolipoma)과 같은 질환에 관여합니다. 대체 스플라이싱으로 인해 세 가지 다른 isoform의 전사 변이가 존재합니다.
