Target Information
RNF168 is a chromatin-associated RING finger protein functioning as a ubiquitin ligase both in vitro and in vivo. It targets histones H2A and H2AX (not H2B), forming K63-linked polyubiquitin chains. Upon DNA double-strand breaks, RNF168 is recruited to damage sites where it co-localizes with γH2AX and 53BP1 in an RNF8-dependent manner. This process enhances the accumulation of ubiquitinated proteins necessary for the retention of 53BP1 and BRCA1, supporting DNA damage response signaling. RNF168 thus plays a critical role in genome maintenance, with at least three known isoforms.
