Target Information
Caspase 8 (CASP8)는 cysteine-aspartic acid protease (caspase) 계열에 속하며, 세포 사멸(apoptosis)의 실행 단계에서 핵심적인 역할을 합니다. Caspase는 비활성 전구체 형태로 존재하며, 내부 아스파르트산 잔기에서 절단되어 활성화됩니다.
Caspase 8은 Fas 및 다양한 세포 사멸 자극에 의해 유도된 programmed cell death에 관여하며, FADD 단백질과 상호작용하는 N-말단 death effector domain을 가집니다.
Huntington 병 환자의 뇌에서 Caspase 8이 불용성 분획에서 검출되었으며, 이는 신경퇴행성 질환과의 관련성을 시사합니다.
활성화된 Caspase 8은 Caspase 9 등 다른 하위 caspase를 활성화하여 세포 사멸을 유도합니다. 또한 Jurkat 세포 및 편도 조직에서도 반응합니다.
Caspase 8의 과발현은 apoptosis를 유도하며, ICE 계열 억제제에 의해 차단될 수 있습니다. 다양한 스플라이싱 변이체가 보고되어 있으나, 모든 변이체의 전체 서열이 확인된 것은 아닙니다.
