Target Information
MCL1 (Myeloid cell leukemia-1) belongs to the Bcl-2 family and is involved in cell differentiation and maintenance of viability, but not proliferation. Isoform 1 inhibits apoptosis while isoform 2 promotes it. The carboxy terminal of MCL1 and Bcl-2 share significant sequence homology. Expression of MCL1 increases upon exposure to DNA damaging agents such as ionizing radiation, ultraviolet radiation, and alkylating drugs.
Enhanced expression of MCL1 is associated with mitochondria and complements Bcl-2 expression in differentiating ML-1 cells. MCL1 promotes cell viability under apoptotic conditions, potentially by neutralizing pro-apoptotic proteins like Bim or Bak. It is expressed in multiple cell types and is essential for embryogenesis and lymphocyte development. MCL1 exists as two isoforms, MCL1L and MCL1S; overexpression of MCL1S promotes cell death.
