Target Information

The androgen receptor (AR) is a member of the steroid-hormone receptor superfamily of nuclear receptors. It is a ligand-activated transcription factor that binds active testosterone (T) and dihydrotestosterone (DHT). Upon ligand binding, the receptor dissociates from accessory proteins, translocates into the nucleus, dimerizes, and stimulates transcription of androgen-responsive genes.

AR signaling is crucial for the development and function of male reproductive organs such as the prostate and epididymis, and also affects nonreproductive organs like muscle, hair follicles, and brain. The receptor is a phosphoprotein that regulates MAP kinase activity, with MEK1/2 inhibition linked to changes in AR phosphorylation. Abnormalities in AR signaling are associated with prostate cancer, Kennedy’s disease, and male infertility. Mutations in the AR gene can cause complete androgen insensitivity syndrome (CAIS).
