Product Specific Information

The SIDI8BEE monoclonal antibody reacts with human and rhesus macaque CD8 beta.
CD8β associates with CD8α to form the CD8αβ heterodimer, expressed on most thymocytes and peripheral cytotoxic αβ TCR T cells. The CD8 receptor complex binds to MHC class I, playing a role in T cell development and activation.

SIDI8BEE detects CD8β in both the CD8αβ heterodimer and CD8ββ homodimer. It is cross-reactive with human and rhesus macaque CD8β (other primates not tested).

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested by flow cytometry on normal human peripheral blood cells.
Use 5 µL (0.2 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Note:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for best results.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining with NovaFluor conjugates.
Add 5 µL per sample to antibody cocktails before combining with cells.
For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology with engineered nucleic acid dye structures that minimize spillover and spread.
Excitation: 636 nm; Emission: 727 nm; Laser: 633–640 nm (Red).

Target Information

The CD8B antigen is a cell surface glycoprotein found on most cytotoxic T lymphocytes.
It mediates cell-cell interactions and acts as a coreceptor with the T-cell receptor (TCR) to recognize antigens presented by MHC class I molecules.
The functional CD8 coreceptor can be a homodimer (αα) or heterodimer (αβ).
Both chains share homology with immunoglobulin light chains.
The CD8B gene encodes beta chain isoforms, with multiple alternatively spliced variants and a pseudogene located on chromosome 2.
