Assay by immunoelectrophoresis resulted in a single precipitin arc against anti-Rabbit Serum, anti-Biotin, and KLH.

Target Information

Keyhole limpet hemocyanin (KLH) is an extremely large, heterogeneous glycosylated protein consisting of subunits with molecular weights of 350,000 and 390,000, forming aggregates of 4,500,000–13,000,000 Da. Each domain of a KLH subunit contains two copper atoms that together bind a single oxygen molecule (O₂). When oxygen is bound, the molecule exhibits a transparent, opalescent blue color.

KLH is highly immunogenic yet safe in humans, making it an ideal vaccine carrier protein. It cannot be reproduced synthetically and is purified from the Keyhole Limpet (Megathura crenulata). KLH is widely used as a carrier protein in research, biotechnology, and therapeutic applications, particularly for coupling haptens such as peptides, small proteins, or drug molecules that are otherwise non-immunogenic.

Its large size, numerous epitopes, and abundant lysine residues allow for high hapten-to-carrier ratios, enhancing hapten-specific antibody generation. As KLH originates from a gastropod, it is phylogenetically distant from mammalian proteins, reducing cross-reactivity in mammalian systems. However, KLH tends to aggregate, which, while still immunogenic, can complicate laboratory handling and conjugation processes.
