Product Specific Information

Description:
The 93 monoclonal antibody reacts with an epitope shared by mouse CD16 and CD32.
CD16 (FcγIII Receptor) and CD32 (FcγII Receptor) are low-affinity receptors for the mouse IgG Fc portion, expressed by B cells, monocytes/macrophages, NK cells, and neutrophils.

Applications Reported:
This antibody has been reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes. Recommended usage ≤0.125 µg/test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test. Titrate for optimal performance.

Fluorophore Information:
PerCP-eFluor® 710 emits at 710 nm and is excited with the blue laser (488 nm).
Recommended filter: 710/50 bandpass (alternative: 695/40 bandpass).
Ensure instrument compatibility.

Fixation:
Samples may be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark with minimal brightness loss.

Excitation/Emission/Laser:
488 nm / 710 nm / Blue Laser

Filtration:
0.2 µm post-manufacturing filtered.

Target Information

CD16:

• 50–65 kDa cell surface molecule
• Exists in transmembrane (NK cells, some T cells) and phosphatidylinositol-linked (granulocytes) forms
• Low-affinity receptor for IgG (FcRIII)
• Mediates ADCC by NK cells

CD32:

• 40 kDa glycoprotein (FcγRII)
• Functions: endocytosis, secretion, cytotoxicity, immunomodulation
• Expressed by B cells, monocytes, granulocytes, platelets
• Involved in phagocytosis of immune complexes and regulation of antibody production
• Genetic variations may increase susceptibility to SLE
