Product Specific Information
The eBioCB16 monoclonal antibody recognizes CD16 (Fc gammaRIII), the low-affinity receptor for IgG (50–80 kDa).
CD16 includes two genes:
- CD16A (FcγRIIIA): expressed in macrophages and NK cells (polypeptide-anchored)
- CD16B (FcγRIIIB): expressed in neutrophils (GPI-anchored)
Two polymorphisms (NA-1, NA-2) exist in CD16B; NA-2 homozygotes show lower phagocytic capacity.
CD16 binds IgG immune complexes, preferentially IgG1 and IgG3, initiating signal transduction cascades that lead to:
- Antibody-dependent cell-mediated cytotoxicity (ADCC)
- Phagocytosis
- Degranulation
- Proliferation
Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Pre-titrated and validated using normal human peripheral blood cells.
Use at 5 µL (0.06 µg) per test (100 µL final volume; 10⁵–10⁸ cells/test).
Fluorochrome Information:
PerCP-eFluor® 710 emits at 710 nm, excited by 488 nm (blue laser).
Recommended filter: 710/50 bandpass (695/40 acceptable alternative).
Light-sensitive; protect from light.
Fixation:
Samples can be stored up to 3 days in the dark at 4°C using:
- IC Fixation Buffer (Product #00-8222)

CD16 (FCGR3A) encodes a receptor that recognizes the Fc portion of IgG, mediating:

Gene Information:
- FCGR3A and FCGR3B share high similarity (chromosome 1)
- FCGR3A: expressed in NK cells (transmembrane anchoring)

Mutations linked to immunodeficiency 20, systemic lupus erythematosus, recurrent viral infections, and alloimmune neonatal neutropenia.
