Product Specific Information

The M1/70 monoclonal antibody reacts with mouse CD11b, a 165–170 kDa integrin alphaM that forms the alphaMbeta2 integrin (Mac-1) with CD18. CD11b binds to CD54 (ICAM-1), C3bi, and fibrinogen, and is expressed on macrophages, NK cells, granulocytes, activated lymphocytes, and B-1 cells in the peritoneal cavity.
M1/70 cross-reacts with human CD11b and can detect this antigen on human peripheral blood monocytes, granulocytes, and a subset of NK cells.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis.
Applications Tested: Mouse splenocytes and bone marrow cells ≤ 0.4 µg/test (100 µL final volume). Cell count: 10⁵–10⁸ cells/test. Optimize by titration.

Compatibility:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01):

• Required for optimal NovaFluor staining.
• Add 5 µL/sample to antibody cocktails before combining with cells.
• For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells).

Technology:
NovaFluor conjugates use Phiton™ technology with engineered nucleic acid dye structures for optimized fluorescent signatures minimizing spillover and spread.
Excitation: 568 nm; Emission: 568 nm; Laser: 561 nm (Yellow).

Note:
Internal testing indicates NovaFluor Yellow 570 non-specifically stains B cells in SJL mice but not in BALB/c or C57BL/6 mice. Other strains untested.

Target Information

CD11b (integrin alpha-M, ITGAM) is a 165 kDa adhesion molecule forming a heterodimer with CD18 (Mac-1, CR3). Expressed on monocytes/macrophages, granulocytes, activated lymphocytes, NK cells, dendritic cells, and microglia.
Functions as a receptor for ICAM-1, ICAM-2, ICAM-4, CD14, CD50, CD23, heparin, iC3b, fibrinogen, and Factor X — critical for cell-cell and cell-matrix interactions.
Expression levels vary by tissue and maturation stage. Associated with systemic lupus erythematosus and ITGAM-related susceptibility to autoimmune disease.
