Product Specific Information

The M1/70 monoclonal antibody reacts with mouse CD11b (integrin alphaM, 165–170 kDa). CD11b associates with CD18 to form Mac-1 (αMβ2 integrin), binding to CD54 (ICAM-1), C3bi, and fibrinogen.
Mac-1 is expressed by macrophages, NK cells, granulocytes, activated lymphocytes, and B-1 cells in mouse peritoneal cavity.
M1/70 also cross-reacts with human CD11b, enabling detection on human monocytes, granulocytes, and NK cell subsets.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Flow cytometric analysis of mouse splenocytes and bone marrow cells (≤ 0.4 µg/test).
A test is defined as antibody amount staining 10⁵–10⁸ cells in 100 µL final volume. Titrate for optimal performance.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining.
  • Add 5 µL per sample (regardless of NovaFluor count).
  • For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells).

Technology:
NovaFluor conjugates use Phiton™ technology, based on nucleic acid dye structures engineered for optimized fluorescence signatures minimizing spillover and spread.

Note:
Internal testing shows NovaFluor Yellow 590 may non-specifically stain B cells in SJL mice. No non-specific staining observed in BALB/c or C57BL/6 mice.

CD11b (Integrin αM, ITGAM)

• 165 kDa adhesion molecule forming heterodimer with CD18 (β2 integrin).
• Known as Mac-1 or CR3 (Complement Receptor 3).
• Expressed on monocytes/macrophages, granulocytes, activated lymphocytes, NK cells, dendritic cells, and microglia.
• Binds ICAM-1/2/4, CD14, CD50, CD23, heparin, iC3b, fibrinogen, Factor X.
• Involved in cell-cell and cell-matrix interactions.
• Expression levels vary by tissue and maturation stage.
• Commonly used as microglial marker.
• Associated with systemic lupus erythematosus and ITGAM-related susceptibility.
