MA1-750 detects prion protein (PrP) from species possessing the conserved n-IHFG-c epitope, including sheep, bovine, deer, and elk.
It has been validated for Western blot, immunohistochemistry, and ELISA applications.
By Western blot, the antibody detects a 33–35 kDa protein in normal samples and a 27–30 kDa PrP fragment in brain protease-treated extracts from infected animals.
In immunohistochemistry, MA1-750 specifically stains PrP(Sc) in infected sheep brain tissue, showing intense staining of spongiform lesions, while not detecting PrP(C).
The immunogen peptide (Cat. # PEP-051) corresponding to residues S(146)–R(159) of bovine PrP is available for neutralization and control experiments.
MA1-750 specifically recognizes a conserved epitope of the PrP(Sc) protein comprising the amino acids n-IHFG-c.

Target Information

Human prion protein (PrP), also known as PRNP, is a GPI-anchored cell surface glycoprotein associated with lipid rafts and functions as a signaling molecule.
PrP plays roles in apoptosis, epithelial cell proliferation, and junction-associated protein distribution in enterocytes.
Conversion of normal PrPc to abnormal PrPSc is the key event in prion neurodegenerative diseases such as Creutzfeld-Jakob disease (CJD).
PrPc is rich in alpha-helices, while PrPSc contains more beta-sheets and is resistant to proteinase K.
