Target Information
NLK (Nemo-like kinase)는 세린/트레오닌 키나아제로, 세포 신호전달 및 세포 간 커뮤니케이션에 관여합니다.
비정형 Wnt 신호전달 경로의 양성 조절자로 작용하며, WNT5A, MAP3K7/TAK1, HIPK2 하위에서 작용합니다.
이 경로의 활성화는 히스톤 메틸트랜스퍼레이스 SETDB1과의 결합 및 인산화를 유도하고, NLK-SETDB1 복합체는 PPARG와 상호작용하여 히스톤 H3K9 메틸화를 통한 전사 억제를 유도합니다.
이로 인해 지방세포 분화를 억제하고 골아세포 분화를 촉진합니다.
또한 canonical Wnt/β-catenin 및 Notch 신호전달 경로의 음성 조절자로 작용하여, TCF7L2/LEF1 복합체 및 NOTCH1 인산화를 통해 전사 활성화를 억제합니다.
MYB 전사인자 패밀리의 음성 조절자 역할도 수행합니다.
