• Detects expression of C-terminal His fusion proteins from bacterial, insect, and mammalian cells.
• Contains enough material for 25 Western blots.
• Functionally tested using 500 ng of E. coli–expressed fusion protein containing a C-terminal polyhistidine (6xHis) epitope.
• Detectable signal observed with 100 ng of recombinant Positope™ protein.
• For Western blot, dilute in PBS with 0.05% Tween-20 and 5% nonfat dry milk (PBSTM).
• Successfully used for immunoprecipitation of 6x His-tagged fusion proteins.

Target Information

The 6x His tag is a synthetic oligopeptide consisting of six consecutive histidine residues (HHHHHH). It is commonly expressed at either the N- or C-terminal of recombinant proteins to facilitate purification via immobilized metal affinity chromatography (IMAC).

Epitope tagging allows detection of recombinant proteins lacking specific antibodies by fusing known epitopes. His-tagged proteins bind strongly and selectively to Ni²⁺-NTA adsorbents, enabling purification to homogeneity. His tag-specific antibodies are useful for detection and co-immunoprecipitation of tagged proteins.
