Target Information

CD20 is a non-glycosylated surface phosphoprotein with a molecular weight range of 33–37 kDa depending on phosphorylation. It is expressed on mature and most malignant B cells, as well as a subset of T lymphocytes and follicular dendritic cells. CD20 expression coincides with surface IgM and regulates calcium conductance, cell cycle progression, and B-cell proliferation.
CD20 is associated with lipid rafts, with its intensity influenced by extracellular triggers and BCR aggregation. Upon receptor ligation, CD20 and BCR colocalize and then dissociate before BCR endocytosis, while CD20 remains at the cell surface.
CD20 is a key therapeutic target for antibody-mediated B-cell depletion, as it is highly expressed in B-cell malignancies and remains stable on the plasma membrane after treatment.
Diseases associated with CD20 dysfunction include Ms4a1-related common variable immune deficiency.
