The 17B5 monoclonal antibody reacts with mouse CD137 (4-1BB), a TNFR superfamily member expressed on activated T cells around day 4–6 after TCR activation. CD137 interaction with its ligand 4-1BBL delivers a costimulatory signal crucial for T cell–APC interaction. The 17B5 antibody shows strong antagonistic effects in functional assays.
Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Validated by flow cytometric analysis of activated mouse splenocytes at ≤0.5 µg/test (100 µL final volume). Optimal performance requires empirical titration (10⁵–10⁸ cells/test).
Target Information
CD137 (TNFRSF9, 4-1BB) is an inducible costimulatory molecule expressed primarily on activated T cells. Its ligand, 4-1BBL, is found on activated macrophages, mature B cells, hematopoietic stem cells, and myeloid progenitor cells. CD137 signaling supports activated and memory CD8⁺ T cell survival and proliferation, while modulating myelopoiesis and dendritic cell development. Soluble CD137 may act as a negative regulator of immune responses.
