Product Specific Information

The 145-2C11 monoclonal antibody reacts with mouse CD3e, a 20 kDa subunit of the TCR complex. The epsilon chain is required for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes and mature T cells. Binding of 145-2C11 to TCR can induce activation, proliferation, or apoptosis depending on conditions. Commonly used as a phenotypic marker for mouse T cells.

  • Applications Tested: Tested by flow cytometric analysis of mouse splenocytes (≤1.0 µg/test).
    A test is defined as the amount of antibody that stains a 100 µL cell sample. Cell number can range from 10⁵–10⁸ cells/test. Titrate for optimal performance.

Fluorochrome Information:
Brilliant Violet™ 786 (BV786) emits at 786 nm and is suitable for cytometers with a 405 nm violet laser. Ensure instrument compatibility.

Buffer Recommendations:
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (SB-4401-42) or Brilliant Stain Buffer™ (00-4409-75) to minimize non-specific interactions.

Light Sensitivity:
Protect from light due to photo-induced oxidation sensitivity.

Fixation:
Samples can be stored in IC Fixation Buffer (00-8222-49) or 1-step Fix/Lyse Solution (00-5333-54) up to 3 days at 4°C in the dark. BV786 is not compatible with methanol-based fixation.

Target Information

The CD3 subunit complex transduces antigen-recognition signals into T cells and regulates TCR complex expression. CD3 subunits (gamma, delta, epsilon, zeta) are immunoglobulin superfamily members encoded on chromosome 11. They associate with cytoplasmic signaling molecules via double tyrosine motifs. CD3 is expressed on 68–82% of peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells, but not on B or NK cells. Defects in CD3 genes can lead to CD3 immunodeficiency.
