Product Specific Information

The OKT8 monoclonal antibody reacts with the human CD8a molecule, a 32–34 kDa cell surface receptor expressed as a heterodimer (CD8αβ) or homodimer (CD8αα). CD8a is found on most thymocytes and subsets of mature T and NK cells. It binds MHC class I and associates with p56lck, influencing T-cell development and activation.

Preliminary testing shows OKT8, RPA-T8, and HIT8a clones do not compete for binding to human peripheral blood leukocytes, suggesting distinct epitopes.

Each product includes 1 vial of NovaFluor conjugate and 1 vial of CellBlox Plus Blocking Buffer.

  • Applications Reported: Flow cytometric analysis
  • Applications Tested: Pre-titrated and validated for flow cytometry (5 µL (0.8 µg)/test)
    • A test = antibody amount for 100 µL staining volume
    • Cell range: 10⁵–10⁸ cells/test

Important Notes:

  • NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
  • Use CellBlox Plus Blocking Buffer (Product # C001T06F01) for optimal staining.
    • Add 5 µL per sample before mixing with cells
    • For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells)

Target Information

CD8 (Cluster of Differentiation 8) is a type I transmembrane glycoprotein of the immunoglobulin receptor family, essential for T-cell receptor (TCR) co-receptor function with MHC class I molecules. It is primarily expressed on cytotoxic T cells, NK cells, monocytes, and dendritic cells, existing as CD8αβ heterodimers or CD8αα homodimers. CD8 engagement triggers Lck-mediated signaling, leading to lymphokine production and cytotoxic T lymphocyte (CTL) activation, critical for pathogen and tumor clearance. Differentiation of naive CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
