Product Specific Information

The OKT8 monoclonal antibody reacts with human CD8a, a 32–34 kDa cell surface receptor expressed as CD8αβ or CD8αα dimers. It is found on thymocytes, subsets of mature T cells, and NK cells. CD8 binds to MHC class I and associates with p56lck, playing a key role in T-cell development and activation.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells. Recommended use: 5 µL (0.8 µg)/test in 100 µL final volume. Cell number range: 10⁵–10⁸ cells/test.

Important Notes:

• NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
• CellBlox Plus Blocking Buffer (Product # C001T06F01) is required for optimal staining. Add 5 µL per sample before combining with cells.
• For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Target Information

CD8 (Cluster of Differentiation 8) is a type I transmembrane glycoprotein of the immunoglobulin receptor family. It functions as a co-receptor for MHC class I molecules and plays a critical role in T-cell signal transduction, differentiation, and activation. CD8 is expressed mainly on cytotoxic T cells as CD8αβ heterodimers or CD8αα homodimers, and also on subsets of NK cells, monocytes, and dendritic cells. Upon MHC-I/peptide binding, CD8 recruits Lck kinase, initiating CTL activation and immune response against pathogens and tumors. Differentiation of naive CD8⁺ T cells into cytotoxic T lymphocytes is enhanced by IL-2, IL-12, and TGF-β1.
