Product Specific Information

The OKT8 monoclonal antibody reacts with the human CD8a molecule (32–34 kDa), expressed as CD8αβ heterodimer or CD8αα homodimer. It is found on most thymocytes, a subset of mature T cells, and NK cells. CD8 binds to MHC class I and associates with protein tyrosine kinase p56lck, playing a role in T-cell development and activation.

OKT8, RPA-T8, and HIT8a clones do not compete for binding to human peripheral blood leukocytes, suggesting distinct epitope recognition.

Each product includes:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated for flow cytometry using normal human peripheral blood cells.
Recommended use: 5 µL (0.5 µg)/test in 100 µL final volume (10⁵–10⁸ cells/test).

Important Notes:

  • NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
  • CellBlox Plus Blocking Buffer (Product # C001T06F01) must be used for optimal staining.
    • Add 5 µL per sample to antibody cocktails before combining with cells.
    • For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, based on novel nucleic acid dye structures engineered for optimized fluorescence with minimal spillover and spread.

Target Information

CD8 (Cluster of Differentiation 8) is a type I transmembrane glycoprotein of the immunoglobulin receptor family. It functions as a co-receptor with TCR for MHC class I molecules, facilitating T cell activation and differentiation.
CD8 is expressed on cytotoxic T lymphocytes, NK cells, monocytes, and dendritic cells as either CD8αβ heterodimers or CD8αα homodimers.
Upon MHC-I/peptide complex binding, CD8 recruits Lck kinase, leading to activation of cytotoxic T lymphocytes (CTLs) involved in pathogen and tumor cell clearance.
Differentiation of naïve CD8⁺ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
