Product Specific Information
The OKT8 monoclonal antibody reacts with the human CD8a molecule, a 32–34 kDa cell surface receptor expressed either as a heterodimer (CD8αβ) or homodimer (CD8αα). CD8a is found on thymocytes, mature T cells, and NK cells. It binds to MHC class I and associates with protein tyrosine kinase p56lck, playing a role in T-cell development and activation.
OKT8, RPA-T8, and HIT8a clones do not compete for binding on human peripheral blood leukocytes, indicating distinct epitopes.
Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated for 4 µL (0.5 µg) per test (100 µL final volume). Recommended cell range: 10⁵–10⁸ cells/test.
Note: NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
Each NovaFluor conjugate ships with CellBlox Blocking Buffer.
Add 5 µL per sample when staining with NovaFluor conjugates, including single-color controls.

Target Information
Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein of the immunoglobulin receptor family. It functions as a co-receptor with TCR for MHC class I molecules. CD8 is primarily expressed on T cells as CD8αβ heterodimers and on subsets of T cells, NK cells, monocytes, and dendritic cells as CD8αα homodimers.
CD8 ligation with MHC-I/peptide complexes recruits Lck kinase, triggering lymphokine production, motility, and cytotoxic T lymphocyte (CTL) activation. Activated CTLs are essential for pathogen and tumor clearance. Differentiation of naive CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
