Product Specific Information
This antibody is predicted to react with bovine, chicken, canine, amphibian, mouse, and rat based on sequence homology.
Heat-mediated antigen retrieval is recommended prior to staining, using a 10 mM citrate buffer (pH 6.0) for 10 minutes followed by cooling at room temperature for 20 minutes.
Following antigen retrieval, incubate samples with primary antibody for 10 minutes at room temperature.
A suggested positive control is breast carcinoma.
Target Information
GRP78 is a 78 kDa glucose-regulated protein belonging to the heat shock protein family (HSP70). It resides in the endoplasmic reticulum (ER) and transiently associates with newly synthesized secretory and membrane proteins.
It interacts with mutant and misfolded proteins, marking them for degradation. The conserved Lys-Asp-Glu-Leu (KDEL) sequence at its C-terminus ensures ER retention via the KDEL receptor.
GRP78 is essential for protein folding and degradation, maintaining cell homeostasis, and preventing apoptosis.
It serves as a biomarker for hypoglycemia and has neuroprotective roles under oxidative stress. Reduced GRP78 levels are observed in Alzheimer’s disease and presenilin-1 mutations.
Induction of GRP78 has been associated with drug resistance to antitumor agents.
