Product Specific Information

  • Immunohistochemistry (PFA fixed): incubate antibody for 30 minutes at room temperature.
  • Staining of formalin-fixed tissues requires heating sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

Fibronectin is a disulfide-bonded dimer with a molecular weight range of 230–250 kDa. In the extracellular matrix of connective tissues and vessels, fibronectin exists as an insoluble protein cross-linked by interchain disulfide bonds forming high molecular mass polymers. It is abundant during embryonic development and tissue remodeling and also found as a soluble plasma protein.

Fibronectin forms repeating modules that create domains for interactions with itself, other matrix components (e.g., collagen, heparin), and cell receptors (e.g., integrins). It plays roles in cell adhesion and migration processes including embryogenesis, wound healing, blood coagulation, host defense, and metastasis. Increased fibronectin expression is associated with non-small cell lung carcinoma and contributes to tumorigenicity and chemoresistance.

The fibronectin gene has three regions subject to alternative splicing, potentially producing up to 20 transcript variants, though the full-length nature of some variants remains undetermined.
