Target Information

Phosphoinositide-specific phospholipase C (PLC) plays a crucial role in receptor-mediated signal transduction through the generation of two second messengers: inositol 1,4,5-triphosphate and diacylglycerol from phosphatidylinositol 4,5-bisphosphate. Mammalian PLC isozymes include PLCβ1, PLCβ2, PLCβ3, PLCβ4, PLCγ1, PLCγ2, PLCδ1, PLCδ2, and PLCε.

PLCδ exists as four isoforms. PLCδ1, a calcium signal amplifier, is activated by an atypical GTP-binding protein and acts as an effector for transglutaminase II-mediated oxytocin receptor and α1B-adrenoreceptor signaling. Mouse PLCδ1 is highly expressed in brain, heart, lung, and testis. Abnormal accumulation of PLCδ in Alzheimer’s disease brains suggests its role in AD pathology. PLCδ2 is markedly expressed in type II intestinal metaplasia and adenocarcinoma, and its expression may serve as a marker of gastric malignant transformation.
