Target Information
Phosphoinositide-specific phospholipase C (PLC)는 수용체 매개 신호전달의 시작에 중요한 역할을 하며, phosphatidylinositol 4,5-bisphosphate로부터 두 가지 2차 전달물질인 inositol 1,4,5-triphosphate와 diacylglycerol을 생성합니다.
PLC는 다양한 이소형(PLCβ1, PLCβ2, PLCβ3, PLCβ4, PLCγ1, PLCγ2, PLCδ1, PLCδ2, PLCε)을 포함하며, PLCδ는 4가지 isoform으로 존재합니다. PLCδ1은 칼슘 신호 증폭기로 작용하며 비정형 GTP 결합 단백질에 의해 활성화됩니다. 또한 PLCδ1은 transglutaminase II 매개 oxytocin receptor 및 α1B-adrenoreceptor 신호전달의 effector로 작용합니다.
Mouse PLCδ1은 뇌, 심장, 폐, 고환에서 높게 발현되며, Alzheimer’s disease(AD) 환자의 뇌에서 비정상적으로 축적되어 AD 병리와 관련이 있을 수 있습니다. PLCδ2는 type II intestinal metaplasia 및 adenocarcinoma에서 강하게 발현되며, gastric malignant transformation의 잠재적 마커로 고려됩니다.
