Preparation:
Produced in BALB/c mice immunized with highly pure Recombinant Human IFN-gamma. Anti-Human IFN-gamma-specific antibody was purified from cell culture by Protein A affinity chromatography.

Sandwich ELISA:
Assuming 100 µL/well, a concentration of 4.0–8.0 µg/mL of this antibody will detect at least 500 pg/mL of Recombinant Human IFN-gamma when used with PeproTech Biotinylated Antigen Affinity Purified Anti-Human IFN-gamma (500-P32BT) as the detection antibody at approximately 2.0–4.0 µg/mL.

Western Blot:
To detect Human IFN-gamma by Western Blot, use at 0.20–0.40 µg/mL. Detection limit for Recombinant Human IFN-gamma is 1.0–2.0 ng/lane under reducing or non-reducing conditions.

IFN gamma (Interferon gamma, Type II interferon)
IFN gamma is a macrophage activation factor produced primarily by T-lymphocytes and NK cells in response to antigens, mitogens, and cytokines. It is a dimeric glycoprotein (~45 kDa) composed of two 146 amino acid subunits.
On SDS-PAGE, it appears as 25, 20, and minor 15.5 kDa bands due to differential glycosylation.
The biological activity is species-specific; human IFN gamma does not cross-react with mouse IFN gamma.

Functions include:

• Antiviral and tumor antiproliferative activity
• Induction of class I and II MHC
• Macrophage activation
• Enhanced immunoglobulin secretion by B lymphocytes
• Cytokine regulation and synergy with other cytokines

Activation occurs through binding to IFN gamma receptor I and II and activation of the JAK-STAT pathway.
Human IFN gamma shares about 40% sequence homology with mouse IFN gamma.
It is upregulated by IL2, FGF basic, and EGF, and downregulated by vitamin D3 or DMN.
Mutations in the IFN gamma gene are associated with aplastic anemia.
