Product Specific Information
The V3GYWCH monoclonal antibody reacts with human and mouse interferon regulatory factor 8 (IRF8; ICSBP). IRF8 is a 50 kDa transcription factor essential for the development of dendritic cells (DC), particularly plasmacytoid DC (pDC) and CD8α⁺ DC. IRF8⁻/⁻ mice lack both pDC and CD8α⁺ DC populations. Although CD8α⁻ DC are present, they fail to mature upon Toll-like receptor signaling and are functionally impaired. Retroviral IRF8 transduction can restore pDC development from DC progenitors in IRF8⁻/⁻ mice.
Applications Reported: Flow cytometric analysis
Applications Tested: Tested by flow cytometric analysis of normal human peripheral blood cells. Recommended ≤ 0.25 µg/test (100 µL final volume). Cell number should be optimized empirically (10⁵–10⁸ cells/test).
Excitation: 488 nm
Emission: 519 nm
Laser: Blue Laser
Target Information
IRF8 (interferon regulatory factor 8) functions as a transcriptional activator or repressor, binding to the upstream regulatory region of type I IFN and IFN-inducible MHC class I genes. It plays a key negative regulatory role in immune cells and is involved in CD8⁺ dendritic cell differentiation through complex formation with the BATF-JUNB heterodimer. This interaction recognizes the AICE sequence, leading to cooperative gene activation. Mutations in IRF8 can cause immunodeficiency 32A and 32B.
