Product Specific Information

This monoclonal antibody 9A11-CD33 reacts with mouse CD33 (Siglec3, gp67). CD33 is part of the sialic acid-binding immunoglobulin lectins (Siglec) family, binding sialic acid-containing glycans on mammalian cell surface glycoproteins and glycolipids. Mouse CD33 is expressed by mature granulocytes, immature myeloid cells, and microglia. Unlike human CD33, mouse CD33 lacks a cytoplasmic ITIM domain and exists as two splice variants with different cytoplasmic tails. In microglia, CD33 inhibits phagocytosis of toxic amyloid beta.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes at ≤ 0.25 µg/test (100 µL final volume). Optimal titration recommended.

Fluorochrome Information:
Super Bright 702 is a tandem dye excited by the violet laser line (405 nm) and emits at 702 nm. Recommended filter: 710/50 bandpass. Ensure instrument compatibility.

Staining Buffer Recommendation:
Use Super Bright Complete Staining Buffer (Product # SB-4401) when using multiple Super Bright dyes to minimize non-specific polymer interactions.

Light Sensitivity: Protect vial and stained samples from light.
Fixation: Samples can be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-57) for up to 3 days at 4°C in the dark. Performance may vary by clone.

Target Information

CD33 is a transmembrane protein of the Siglec family with ITIM motifs that recruit SHP-1 and SHP-2 phosphatases. It is expressed on myelomonocytic lineage cells and binds sialic acid residues in N- and O-glycans. CD33 is a therapeutic target for acute myeloid leukemia and is expressed on granulocyte and macrophage precursors, but not on pluripotent stem cells. It serves as a marker for peripheral monocytes and helps distinguish myelogenous leukemia cells from lymphoid or erythroid leukemias. Diseases linked to CD33 dysfunction include gallbladder lymphoma and extracutaneous mastocytoma.
