Product Specific Information

The Ber-H2 monoclonal antibody reacts with human CD30, a 120 kDa transmembrane glycoprotein of the TNFR superfamily. CD30 is expressed on activated T and B lymphocytes but not on resting lymphocytes, and is also found on certain lymphomas, embryonal carcinomas, and virus-infected lymphocytes (HIV, HTLV, EBV). CD30-CD30L interaction activates NF-κB signaling and regulates apoptosis.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested by flow cytometry of normal human peripheral blood cells. Use 5 µL (0.5 µg) per test in a final volume of 100 µL. Cell number range: 10⁵–10⁸ cells/test.

Fluorochrome Information:
Super Bright 702 is a tandem dye excited with a 405 nm violet laser and emits at 702 nm. Recommended filter: 710/50 bandpass. Ensure instrument compatibility.

Staining Recommendations:
When using multiple Super Bright-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize nonspecific polymer interactions.

Light Sensitivity: Protect from light due to photo-induced oxidation.
Fixation: Samples can be fixed with IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-57) for up to 3 days at 4°C in the dark with minimal signal loss.

Target Information

CD30 (Ki-1, TNFRSF8) is a type I transmembrane glycoprotein of the TNF receptor superfamily, originally identified on Hodgkin and Reed-Sternberg cells. CD30 binding to CD30L mediates cell proliferation, differentiation, and apoptosis, playing a key role in Hodgkin’s disease and CD30⁺ lymphomas. It also functions as a costimulatory molecule in thymic selection and is expressed in activated T and B cells, some non-Hodgkin’s lymphomas, and virus-infected lymphocytes. The soluble form (sCD30) serves as a Th2 immune response marker. CD30 interacts with TRAF2 and TRAF5 to activate NF-κB and regulate apoptosis.
