Product Specific Information

The 5C3 monoclonal antibody reacts with human CD40, a 45–50 kDa type I transmembrane glycoprotein belonging to the TNFR family. CD40 is expressed by B lymphocytes, follicular dendritic cells, thymic epithelium, and a subset of peripheral T cells. It plays a key role in B cell development, maturation, and immune regulation by interacting with CD154 (gp39) on T cells.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on normal human peripheral blood cells at ≤5 µL (0.125 µg)/test. Recommended titration for optimal performance.

Fluorochrome Information:
Super Bright 702 is a tandem dye excited by the violet laser (405 nm) and emits at 702 nm. Recommended filter: 710/50 bandpass. Protect from light.

Buffer Recommendations:
When using multiple Super Bright conjugates, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark.

Target Information

CD40 is a single-chain glycoprotein belonging to the TNF/NGF receptor superfamily. It is expressed on B cells (except plasma cells), epithelial cells, carcinomas, and lymphoid dendritic cells. CD40 mediates immune and inflammatory responses including class switching, memory B cell development, and germinal center formation. Dysregulation is associated with Type 3 Hyper-IgM immunodeficiency and CD40 ligand deficiency.
