Target Information

Dihydropyridine receptor (DHPR) is a surface membrane protein critical for excitation-contraction coupling of striated muscle. DHPR and the sarcoplasmic reticulum ryanodine receptor (RyR) are key components of intracellular junctions, converting surface membrane depolarization into Ca²⁺ release from internal stores.
The alpha1-subunit of DHPR contains a cytoplasmic loop involved in interactions with RyR. Phosphorylation of the DHPR alpha1-subunit may influence functional interaction between DHPR and RyR. Mutation in DHPR alpha1 leads to excitation-contraction uncoupling and muscular dysgenesis. Cells lacking RyR exhibit reduced Ca²⁺ current density and absence of excitation-contraction coupling.
