Preparation:
Produced from sera of rabbits immunized with highly pure Recombinant Rat IFN-gamma. Anti-Rat IFN-gamma-specific antibody was purified by affinity chromatography employing an immobilized Rat IFN-gamma matrix.

Sandwich ELISA:
To detect Rat IFN-gamma by sandwich ELISA (using 100 µL/well antibody solution), a concentration of 0.5–2.0 µg/mL of this antibody is required. This antigen affinity purified antibody, in conjunction with PeproTech Biotinylated Anti-Rat IFN-gamma (500-P122Bt) as a detection antibody, allows the detection of at least 0.2–0.4 ng/well of Recombinant Rat IFN-gamma.

Western Blot:
To detect Rat IFN-gamma by Western Blot analysis, this antibody can be used at a concentration of 0.1–0.2 µg/mL. When used with compatible secondary reagents, the detection limit for Recombinant Rat IFN-gamma is 1.5–3.0 ng/lane under either reducing or non-reducing conditions.

IFN gamma (Interferon gamma, Type II interferon)

• Macrophage activation factor produced mainly by T-lymphocytes and natural killer cells.
• Dimeric glycoprotein (~45 kDa) consisting of two 146 amino acid subunits.
• Appears as 25, 20, and minor 15.5 kDa bands on SDS-PAGE due to differential glycosylation.
• Highly species-specific; human IFN gamma does not cross-react with mouse.
• Functions include antiviral activity, tumor antiproliferative activity, induction of class I and II MHC, macrophage activation, and enhanced immunoglobulin secretion by B lymphocytes.
• Acts synergistically with other cytokines and activates the JAK-STAT pathway via IFN gamma receptor I and II.
• Upregulated by IL2, FGF basic, EGF; downregulated by vitamin D3 or DMN.
• Gene mutations associated with aplastic anemia.
