Preparation:
Produced from sera of goats immunized with highly pure Recombinant Rat IFN-gamma. Anti-Rat IFN-gamma specific antibody was purified by affinity chromatography employing an immobilized Rat IFN-gamma matrix.

Sandwich ELISA:
To detect Rat IFN-gamma by sandwich ELISA (using 100 µL/well antibody solution), a concentration of 0.5–2.0 µg/mL is required. This antibody, in conjunction with PeproTech Biotinylated Anti-Rat IFN-gamma (500-P122GBt) as a detection antibody, allows detection of at least 0.2–0.4 ng/well of Recombinant Rat IFN-gamma.

Western Blot:
To detect Rat IFN-gamma by Western Blot analysis, use at a concentration of 0.1–0.2 µg/mL. When used with compatible secondary reagents, the detection limit for Recombinant Rat IFN-gamma is 1.5–3.0 ng/lane under reducing or non-reducing conditions.

IFN gamma (Interferon gamma, Type II interferon)
IFN gamma is a macrophage activation factor and immune interferon produced primarily by T-lymphocytes and natural killer cells in response to antigens, mitogens, and cytokines. It is a dimeric glycoprotein consisting of two 146 amino acid subunits (approx. 45 kDa). On SDS-PAGE, it appears as 25, 20, and minor 15.5 kDa bands due to differential glycosylation.

Biological functions include:

• Antiviral activity
• Tumor antiproliferative activity
• Induction of class I and II MHC
• Macrophage activation
• Enhanced immunoglobulin secretion by B lymphocytes

IFN gamma acts via IFN gamma receptor I and II through the JAK-STAT pathway. Human IFN gamma shows ~40% sequence homology with mouse IFN gamma and does not cross-react with mouse IFN gamma. Expression is upregulated by IL2, FGF basic, EGF and downregulated by vitamin D3 or DMN. Gene mutations are associated with aplastic anemia.
