This antibody detects endogenous levels of IRE1 only when phosphorylated at phospho Y628.
Target Information
ERN1 (serine/threonine kinase) or endoplasmic reticulum to nucleus signaling 1 is a human homologue of the yeast Ire1 gene product. It possesses intrinsic kinase and endoribonuclease activities, playing a key role in ER-based stress signaling. ERN1 controls IRE1 proteolysis in mammalian cells and is essential for fetal viability in extraembryonic cells. IRE1 acts as a sensor of unfolded proteins in the ER, initiating the unfolded protein response by splicing XBP1 mRNA into a potent transcriptional activator. While null mutants are viable, IRE1 is essential under stress conditions that cause unfolded proteins to accumulate in the ER. Depletion of IRE1p has no effect under normal conditions but induces apoptosis under ER stress.
