Heat-mediated antigen retrieval is recommended prior to staining, using a 10 mM citrate buffer (pH 6.0) for 10 minutes, followed by cooling at room temperature for 20 minutes.
After antigen retrieval, incubate samples with primary antibody for 30 minutes at room temperature.
A suggested positive control is mesothelioma tissue.

Target Information

Mesothelin was originally identified as a differentiation antigen of mesotheliomas, ovarian cystadenocarcinomas, and pancreatic adenocarcinomas.
It is a glycosylphosphatidylinositol (GPI)-anchored, cell-surface glycoprotein predominantly secreted by mesothelial cells.
While normal mesothelial cells express Mesothelin at restricted levels, aberrant overexpression is observed in various cancers including endometrioid uterine adenocarcinoma and squamous cell carcinomas of the esophagus, stomach, lung, and cervix.

Proteolytic cleavage of Mesothelin yields a soluble fragment known as megakaryocyte potentiating factor (MPF), which stimulates megakaryocyte colony formation and may regulate platelet production.
High expression of Mesothelin and MPF in cancerous tissues suggests their involvement in tumorigenesis, promoting tumor cell proliferation, migration, and metastasis.
