Arginase-1 (Arg1) is a 35 kDa enzyme that converts L-arginine to urea and L-ornithine, completing the urea cycle. The resulting polyamines are essential for cell proliferation and detoxification of protein degradation byproducts. Arginase 1 also down-regulates nitric oxide production by depleting NO synthase substrate.
In humans and mice, Arg1 is expressed in the liver, neutrophils, myeloid-derived suppressor cells (MDSC), and neural stem cells. In humans, it is found in blood neutrophils but not CCR3+ granulocytes; in mice, it marks alternatively activated macrophages (M2a).
Arginase-1 is often expressed in myeloid cells infiltrating tumors and is typically found in most hepatocellular carcinomas. Defects in Arginase 1 cause argininemia, an autosomal recessive disorder characterized by hyperammonemia.
