Description:
The RPA-T4 monoclonal antibody reacts with human CD4, a 59 kDa cell surface receptor expressed by most thymocytes, a subset of mature T cells (T-helper cells), and at low levels on monocytes. CD4 is a receptor for the human immunodeficiency virus (HIV). RPA-T4 blocks HIV binding and mixed lymphocyte reaction. It recognizes a different epitope than the OKT4 monoclonal antibody, and these antibodies do not cross-block each other’s epitopes.

Applications Reported:
The RPA-T4 antibody is reported for use in flow cytometric analysis and blocking of CD4 activity in in vitro functional assays.

Applications Tested:
Tested by flow cytometric analysis of normal human peripheral blood cells. Recommended ≤1 µg/test. A test is defined as the amount of antibody staining a cell sample in 100 µL final volume. Cell number should be empirically determined (10⁵–10⁸ cells/test). Titrate for optimal performance.

Use in a sterile environment.

Target Information

The CD4 antigen is involved in recognition of MHC class II molecules and acts as a co-receptor for HIV. CD4 is mainly expressed on T-helper lymphocytes, and also on monocytes, macrophages, and dendritic cells. Tissue expression includes thymus, lymph nodes, tonsils, spleen, and certain brain and gut regions. CD4 participates in early T-cell activation via association with the TCR complex and kinase Lck, and may mediate neuronal damage in immune-related CNS diseases. Multiple alternatively spliced transcripts have been identified (RefSeq, July 2017).
