Product Specific Information

The ICRF44 monoclonal antibody reacts with human CD11b, a 165 kDa adhesion molecule. CD11b associates with integrin beta2 (CD18) and is expressed on monocytes, granulocytes, activated lymphocytes, and a subset of NK cells. It acts as a receptor for ICAM family members (CD54, CD102, CD50) and iC3b, crucial in cell-cell and cell-matrix interactions.

Each product includes:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Pre-titrated and tested using normal human peripheral blood cells at 5 µL (0.4 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability testing.

This conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), required for optimal staining.

  • Add 5 µL per sample to antibody cocktails before mixing with cells.
  • For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, based on novel nucleic acid dye structures enabling engineered fluorescent signatures with minimized spillover and spread.

Target Information

CD11b (integrin alpha-M, ITGAM, integrin alpha-X, ITGAX) forms a heterodimer with CD18 (integrin beta-2), known as Mac-1 or CR3. It is expressed on:

  • Monocytes/macrophages
  • Granulocytes
  • Activated lymphocytes
  • NK cells subset
  • Dendritic cells subset
  • Microglia in the brain

CD11b/CD18 acts as a receptor for ICAM-1, ICAM-2, ICAM-4, CD14, CD50, CD23, heparin, iC3b, fibrinogen, and Factor X, mediating essential adhesion processes.
Expression varies by cell type and tissue, commonly used as a microglial marker.
CD11b dysfunction is linked to systemic lupus erythematosus and ITGAM-related disease susceptibility.
