Product Specific Information

The ICRF44 monoclonal antibody reacts with human CD11b, a 165 kDa adhesion molecule associated with integrin beta2 (CD18). CD11b is expressed on monocytes, granulocytes, activated lymphocytes, and a subset of NK cells. It functions as a receptor for ICAM family members (CD54, CD102, CD50) and iC3b, playing a key role in cell-cell and cell-matrix interactions.

Each product includes:

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells. Recommended usage is 5 µL (0.4 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability testing with NovaFluor dyes.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining. Add 5 µL per sample before combining antibodies with cells.
• For single-color controls, use 5 µL of buffer per 100 µL of cell sample.

Target Information

CD11b (integrin alpha-M, ITGAM) associates non-covalently with integrin beta-2 (CD18) to form the CD11b/CD18 complex, also known as Mac-1 or CR3. It is expressed on monocytes/macrophages, granulocytes, activated lymphocytes, NK cells, dendritic cells, and microglia. CD11b/CD18 acts as a receptor for ICAM-1, ICAM-2, ICAM-4, CD14, CD50, CD23, heparin, iC3b, fibrinogen, and Factor X, mediating essential adhesion processes.

Expression patterns:

• 8% of spleen cells
• 44% of bone marrow cells
• <1% of thymocytes
  CD11b serves as a microglial marker and its expression increases during monocyte maturation. Higher levels are found in peritoneal macrophages compared to splenic macrophages. CD11b dysfunction is associated with systemic lupus erythematosus and ITGAM-related susceptibility to autoimmune disease.
