Product Specific Information

The ICRF44 monoclonal antibody reacts with human CD11b, a 165 kDa adhesion molecule associated with integrin beta2 (CD18). CD11b is expressed on monocytes, granulocytes, activated lymphocytes, and a subset of NK cells. It serves as a receptor for intercellular adhesion molecules CD54, CD102, CD50, and iC3b, facilitating cell-cell and cell-matrix interactions.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated for use at 5 µL (0.4 µg) per test in 100 µL final volume. Recommended cell range: 10⁵–10⁸ cells/test.

Important Notes:

• NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining.
  • Add 5 µL per sample (regardless of number of NovaFluors).
  • For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

Target Information

CD11b (integrin alpha-M, ITGAM) forms a heterodimer with integrin beta-2 (CD18), known as Mac-1 or CR3. It is expressed on monocytes/macrophages, granulocytes, activated lymphocytes, NK cells, dendritic cells, and microglia. CD11b/CD18 acts as a receptor for ICAMs, CD14, CD50, CD23, heparin, iC3b, fibrinogen, and Factor X, mediating critical adhesion processes.

Expression varies by tissue:

• 8% of spleen cells
• 44% of bone marrow cells
• <1% of thymocytes

CD11b is a common microglial marker and its expression increases during monocyte maturation. Dysregulation is associated with systemic lupus erythematosus and ITGAM-related susceptibility.
